Molecular characterization of Vibrio species and Raoultella ornithinolytica isolated from some seafood’s sold in port Harcourt, river state

Abstract: Vibrio species are known to cause foodborne infections and have been reported severally to possess virulent genes this makes treatment regimens difficult. The aim of this study is to Characterize Vibrio species from seafood’s using Molecular techniques. A total of 972 Fresh and Parboiled samples of Senilia senilis (Blood Cockles), Crassostrea gasar (Oyster), and Tympanotonus fuscatus (Periwinkles) were purchased from Creek Road, Mile 1, and Mile 3 Market, in Port Harcourt Local Government Area, Rivers State, Nigeria. Vibrio species were isolated using Thio-sulphate- Citrate –Bile- Sucrose (TCBS) agar. The 16s ribosomal RNA region of bacterial isolates was amplified using polymerase chain reaction (PCR). Isolates from seafood sample were identified as V. cholerae, V. paraheamolyticus and Raoultella ornithinolytica. The study established the presence of the CtxB and toxR virulence genes in Vibrio cholerae isolates. The presence of these pathogens in seafood samples is a threat to public health as they can cause gastroenteritis and septicemia when consumed raw or undercooked. The filter feeding nature of oysters and cockles also makes them potential reservoirs of virulent genes, this is worrisome. Therefore, seafood handling procedures and processing must be monitored regularly to ensure the microbial quality of these seafood’s is compliant with established standards.
